Short answer
Semaglutide is a peptide, which normally breaks down in the gut. Oral semaglutide is co-formulated with an absorption enhancer (SNAC) and must be taken on an empty stomach with little water, then a wait before eating. Two US brands use it: Rybelsus, approved in 2019 for type 2 diabetes, and the Wegovy pill, a higher-strength tablet approved in December 2025 for weight management. Side effects match injectable semaglutide: mainly nausea, diarrhoea, vomiting and constipation, plus the same label warnings. Peptide Confessions does not publish dosing, timing, injection or sourcing guidance.

How the tablet gets absorbed
The tablet contains SNAC (salcaprozate sodium), which briefly raises pH around the tablet in the stomach, protecting semaglutide from enzymes and helping it cross the stomach lining. Only a small fraction is absorbed, which is why oral doses are much larger than injected doses and why food, coffee or other pills taken too soon cut absorption. Patients follow the timing rules on their own label.
Pill vs injection
Daily pills avoid needles but require a fasting routine; weekly injections are simpler for some. Warnings are shared across semaglutide: the boxed thyroid C-cell tumour warning, pancreatitis, gallbladder problems and dehydration from vomiting or diarrhoea. Community Q&A
- Is oral semaglutide as effective as injections?
- At the higher Wegovy-pill strength, average weight loss in OASIS 4 was in a similar range to injectable Wegovy. Lower Rybelsus strengths produced less weight loss.
- What are oral semaglutide side effects?
- Mainly nausea, diarrhoea, vomiting and constipation, plus the same label warnings as injectable semaglutide.
- Is Rybelsus the same as the Wegovy pill?
- Same molecule, different strengths and approved uses: Rybelsus for type 2 diabetes, the Wegovy pill for weight management.
- Why must oral semaglutide be taken fasting?
- Food and drink sharply reduce how much semaglutide is absorbed from the tablet.
- Does this page give dosing advice?
- Peptide Confessions does not publish dosing, timing, injection or sourcing guidance.
