Vyleesi prescribing information says to inject at least 45 minutes before anticipated sexual activity, and that the duration of efficacy after each dose is unknown — the optimal window has not been fully characterised. Patients may time use based on how they experience desire effects and adverse reactions such as nausea. Do not administer more than one dose within 24 hours per the label. Mean terminal half-life of bremelanotide after a subcutaneous Vyleesi dose is about 2.7 hours (range about 1.9–4.0 hours).
